New highlights of polyphenols from red wine to counteract ocular degenerative diseases

Abstract

More recently, studies have shown that polyphenols could also prevent or improve vision in patients with ocular diseases and especially age-related macular degeneration (AMD) which is an eye disease characterized by damage to the central part of the retina, the macula, and that affects millions of people worldwide. Despite therapeutic advances thanks to the use of anti-vascular endothelial growth factor (VEGF), many resistance mechanisms have been found to accentuate the visual deficit. In the present study, we explored whether a supplementation based on mediterranean diet associating polyphenols from red wine and omega-3 fatty acids was able to counteract laser-induced choroidal neovascularization (cnv) in mice and moleular mechanism involved in neaovasculariation of amd. We highlight that the formulation, resvega®, composed of dha/epa and resveratrol, significantly reduced cnv in mice and a proteomic approach confirmed that resvega® could counteract the progression of amd through a pleiotropic effect targeting key regulators of neoangiogenesis in retina cells in vivo. These events were associated with an accumulation of resveratrol metabolites within the retina. A molecular analysis revealed that resvega®, inhibited vegf-a secretion in vitro by disrupting the dissociation of the VEGF-R2/CAV-1 complex into rafts and subsequently preventing mapk activation and inhibits the phosphorylation of the inhibitor of NFΚB, IΚB, which can bound NFκB dimers and sequester them in the cytoplasm. Moreover, DNA chip analysis reveals that this combination prevents the interaction between ap-1 and VEGF-A and VEGF-R2 gene promoters. Very interestingly, we highlight that resvega® could prolong the anti-angiogenic effect of avastin®, which is an anti-VEGF agent typically used in clinical practice. Furthermore, recent results have shown that a mixture of red wine extract present same effects to counteract the over secretion of VEGF in human retina cells. Therefore, a supplementation of omega-3/resveratrol or with red wine extract could improve the management or slow the progression of AMD in patients with this condition.

Nouveaux apports de polyphénols du vin rouge pour lutter contre les maladies dégénératives oculaires.

Des études récentes ont montré que les polyphénols pourraient prévenir ou améliorer la vision chez les patients atteints de maladies oculaires et notamment de dégénérescence maculaire liée à l’âge (DMLA) qui est une maladie oculaire caractérisée par des lésions de la partie centrale de la rétine, la macula, et qui touche des millions de personnes dans le monde. Malgré les progrès thérapeutiques grâce à l’utilisation d’anticorps ciblant le facteur de croissance endothélial anti-vasculaire (VEGF), de nombreux mécanismes de résistance accentuent le déficit visuel. Dans la présente étude, nous avons exploré si une supplémentation basée sur un régime méditerranéen associant des polyphénols du vin rouge et des acides gras oméga-3 était capable de contrecarrer la néovascularisation choroïdienne (CNV) induite par laser chez la souris et le mécanisme moléculaire impliqué dans la néovascularisation de la DMLA. Nous mettons ainsi en évidence que la formulation resvega®, composée de DHA/EPA et de resvératrol, un polyphenol de la vigne et du vin, permet de réduire significativement la CNV chez la souris, ce qu’une approche protéomique a confirmé en montrant que le resvega® pouvait contrecarrer la progression de la dmla grâce à un effet pléiotropique ciblant les régulateurs clés de la néoangiogenèse dans les cellules de la rétine in vivo. Ces événements étaient associés à une accumulation de métabolites du resvératrol dans la rétine. Une analyse moléculaire a révélée que le resvega® inhibait la sécrétion de VEGF-A in vitro en perturbant la dissociation du complexe VEGF-R2/CAV-1 en radeaux et en empêchant par la suite l’activation de mapk et inhibait la phosphorylation de l’inhibiteur de NFΚB, IΚB, qui peut dimères NFΚB liés et les séquestrent dans le cytoplasme. De plus, l’analyse dna chip révèle que cette combinaison empêche l’interaction entre AP-1 et les promoteurs des gènes VEGF-A et VEGF-R2. De manière très intéressante, nous démontrons quele resvega® pourrait prolonger l’effet anti-angiogénique d’avastin®, qui est un agent anti-VEGF généralement utilisé en pratique clinique. De plus, des résultats récents ont montré qu’un mélange d’extraits de vin rouge présente les mêmes effets pour contrecarrer la sécrétion excessive de VEGF dans les cellules de la rétine humaine. Ainsi, une supplémentation en oméga-3/resvératrol ou en extrait de vin rouge pourrait améliorer la prise en charge ou ralentir la progression de la DMLA chez les patients atteints de cette pathologie.

Nuevos aspectos destacados de los polifenoles del vino tinto para contrarrestar las enfermedades degenerativas oculares.

Estudios recientes han demostrado que los polifenoles podrían prevenir o mejorar la visión en pacientes con enfermedades oculares, en particular la degeneración macular asociada a la edad (DMAE), que es una enfermedad ocular caracterizada por daños en la parte central de la retina, la mácula, y que afecta a la visión. Millones de personas en todo el mundo. A pesar de los avances terapéuticos mediante el uso de anticuerpos dirigidos al factor de crecimiento endotelial vascular (VEGF), numerosos mecanismos de resistencia acentúan el déficit visual. En el presente estudio, exploramos si la suplementación basada en una dieta mediterránea que combina polifenoles del vino tinto y ácidos grasos omega-3 era capaz de contrarrestar la neovascularización coroidea (CNV) inducida por láser en ratones y el mecanismo molecular implicado en la neovascularización de la dmae. De este modo, demostramos que la formulación resvega®, compuesta de dha/epa y resveratrol, un polifenol procedente de la vid y del vino, permite reducir significativamente la cnv en ratones, lo que un enfoque proteómico ha confirmado al demostrar que resvega® podría contrarrestar la progresión de la amd a través de un efecto pleiotrópico dirigido a reguladores clave de la neoangiogénesis en células de la retina in vivo. Estos eventos se asociaron con una acumulación de metabolitos de resveratrol en la retina. El análisis molecular reveló que resvega® inhibía la secreción de vegf-a in vitro al interrumpir la disociación del complejo VEGF-R2/CAV-1 en balsas y posteriormente prevenir la activación de mapk e inhibir la fosforilación del inhibidor de NFκB, IκB, que puede. Se unen a los dímeros de nfκb y los secuestran en el citoplasma. Además, el análisis de adn chip revela que esta combinación previene la interacción entre ap-1 y los promotores de los genes vegf-a y VEGF-R2. Curiosamente, demostramos que resvega® podría prolongar el efecto antiangiogénico de avastin®, que es un agente anti-VEGF generalmente utilizado en la práctica clínica. Además, resultados recientes han demostrado que una mezcla de extractos de vino tinto exhibe los mismos efectos para contrarrestar la secreción excesiva de VEGF en las células de la retina humana. Así, la suplementación con omega-3/resveratrol o extracto de vino tinto podría mejorar el manejo o frenar la progresión de la dmae en pacientes que padecen esta patología.

What drives Indications of Geographical Origin protection and governance mechanisms in the U.S. and European contexts? A contribution of the social sciences

There are fundamentally two different ways in which indications of geographical origin (igos) can be protected. The us approach favors the pre-existing trademark system through collective marks (cms), while the eu approach favors a maximalist approach via a sui generis system which promotes appellations of origin (aos). A consensus however emerges regarding the fundamental protection of origin against misleading, confusing and dilutive uses. Previous literature discusses these competing igo logics from historical, legal and international trade perspectives. In this paper, we depart from the field of social sciences, in particular from recent advancements in the well-established literature on proximities, in order to provide a reflection on the different logics underpinning the aos and cms systems.

Critical investigation on additions to improve the sensory characteristics of dealcoholized wine

The demand for dealcoholized wine has been progressively increasing in recent years. Moreover, the attention for such products is probably increasing even more. Due to that increasing demand and market awareness the legal authorities are about changing rules for that products. Also, at OIV level, these products are being intensively discussed for certain time. The production of dealcoholized wine bases on wine as initial product. This wine is then reduced by physical methods to an alcohol content of less than 0.5% vol., or in other words, to less than 4g/l of alcohol. There are various technologies are possible for producing dealcoholized wine (Schmitt and Christmann 2019).

Unconventional methods to delve deeper into the influence of temperature and nutrition on Chardonnay wine profiles

Temperature and yeast nutrition profoundly impact wine quality and sensory attributes by modulating yeast aroma production and release during fermentation. While temperature and nitrogen’s individual effects are well-studied, their combined influence, including nutrient type and addition timing, remains underexplored. hence, this study aimed to investigate the simultaneous effects of these factors on fermentation kinetics, aroma production and sensory profile, particularly in a Chardonnay wine production selected as a quite aromatically neutral base.

The socioclimatic dynamics and the table grape production during a long-drought: the case of Brazilian semiarid

In 2022, the area cultivated with grapes in Brazil counted 75 thousand ha. About 1/2 of the grape production is located in rio grande do sul state, in South Brazil. Nonetheless, the northeast region, especially the Sao Francisco River Valley (SFRV), is increasing its area and production, mainly pushed by table grapes. The states of bahia and pernambuco already respond for circa 1/3 of brazilian grape production.

Consumer acceptance of sweet wines produced by stopping fermentation with octanoic and decanoic acids

The use of medium chain fatty acids for arresting the fermentation and producing sweet wines was investigated at industrial level. Doses of 10 mg/l of octanoic or decanoic acid and a combination of 5+5 mg/l octanoic and decanoic acid were used to produce sweet wines of tamaioasa romanească variety in volumes of 3000 l.
