Direct-injection HPLC for simultaneous determination of individual and total polyphenols in red wines: validation of the method

Abstract

Polyphenols are very important compounds of red wines, serving as essential bioactive components and playing an important role in sensory properties. The determination of individual phenolic compounds in red wine is commonly performed by hplc analysis, while the total polyphenols are quantified by spectrophotometric methods, usually by the method of absorbance at 280 nm (index of ribéreau-gayon) or the method of index of folin-ciocalteu. In this work, we pioneeringly proposed a new and fast method for simultaneous determination of individual and total polyphenols in red wines by direct-injection hplc without sample preparation. The quantification of total polyphenols was acquired simply by integration of total peaks throughout the elution period of HPLC analysis for the individual phenolic compounds, and thus omit the unnecessary spectrophotometric process. For validation of the proposed method, a model wine solution system, composed of cyanidin- 3-o-glucoside, epicatechin, and acetaldehyde with proportion of 1: 6: 10 (total polyphenols 4.366 mg/ml), 12% ethanol, 5g/l of tartaric acid and at ph 3.2, was established. During the kinetic reaction in model wine solution which simulates the real wine storage and ageing process, the total polyphenols were periodically monitored by the method of index of riberaux-gayon (absorbance at 280 nm) and the method of index of folin-ciocalteu, as well as the proposed peak integration method. The results have showed that there was no significant difference in total polyphenol content among the three methods (p>0.05), all of which presented good accuracy compared to the known polyphenol content in the model wine solution. Moreover, the feasibility of the proposed peak integration method was also verified by its application to quantification of total polyphenols in various different red wines. The results obtained by the proposed peak integration method were well consistent with those obtained by index of riberaux-gayon and index of folin- ciocalteu (p>0.05), indicating that the proposed method is valid for the determination of total polyphenols in red wine.

Analyse HPLC par injection directe pour le dosage simultané des polyphénols individuels et polyphénols totaux dans les vins rouges: validation de la méthode

Les polyphénols sont des composés très importants des vins rouges, servant de composants bioactifs essentiels et jouant un rôle important dans les propriétés sensorielles. La détermination des composés phénoliques individuels dans le vin rouge est généralement effectuée par analyse HPLC, tandis que les polyphénols totaux sont quantifiés par des méthodes spectrophotométriques, généralement par la méthode de l’absorbance à 280 nm (indice de ribéreau-gayon) ou la méthode de l’indice de folin-ciocalteu. Dans ce travail, nous avons proposé une méthode nouvelle et rapide pour la détermination simultanée des polyphénols individuels et totaux dans les vins rouges par hplc à injection directe sans préparation d’échantillon. La quantification des polyphénols totaux a été obtenue simplement par l’intégration des pics totaux tout au long de la période d’élution de l’analyse hplc pour les composés phénoliques individuels, ce qui permet d’éviter le processus spectrophotométrique inutile. Pour la validation de la méthode proposée, un système modèle de solution de vin, composé de cyanidine- 3-o-glucoside, d’épicatéchine et d’acétaldéhyde dans des proportions de 1 : 6 : 10 (polyphénols totaux 4,366 mg/ml), 12% d’éthanol, 5g/l d’acide tartrique et à un ph de 3,2, a été établi.au cours de la réaction cinétique dans une solution de vin modèle qui simule le processus réel de conservation et de vieillissement du vin, les polyphénols totaux ont été périodiquement contrôlés par la méthode de l’indice de riberaux-gayon (absorbance à 280 nm) et la méthode de l’indice de folin-ciocalteu, ainsi que par la méthode d’intégration des pics proposée. Les résultats ont montré qu’il n’y avait pas de différence significative dans la teneur totale en polyphénols entre les trois méthodes (p>0,05), qui présentaient toutes une bonne précision par rapport à la teneur connue en polyphénols dans la solution de vin modèle. En outre, la faisabilité de la méthode d’intégration des pics proposée a également été vérifiée par son application à la quantification des polyphénols totaux dans différents vins rouges. Les résultats obtenus par la méthode d’intégration des pics proposée étaient bien cohérents avec ceux obtenus par l’indice de riberaux-gayon et l’indice de folin-ciocalteu (p>0,05), ce qui indique que la méthode proposée est valable pour la détermination des polyphénols totaux dans le vin rouge.

Análisis HPLC por inyección directa para la determinación simultánea de polifenoles individuales y polifenoles totales en vinos tintos: validación del método

Los polifenoles son compuestos muy importantes de los vinos tintos, que actúan como componentes bioactivos esenciales y desempeñan un papel importante en las propiedades sensoriales. la determinación de compuestos fenólicos individuales en vino tinto se realiza comúnmente por análisis HPLC, mientras que los polifenoles totales se cuantifican por métodos espectrofotométricos, generalmente por el método de absorbancia a 280 nm (índice de ribéreau-gayon) o el método de índice de folin-ciocalteu. en este trabajo, propusimos de forma pionera un método nuevo y rápido para la determinación simultánea de polifenoles individuales y totales en vinos tintos mediante hplc de inyección directa sin preparación de muestras. la cuantificación de los polifenoles totales se adquirió simplemente por integración de los picos totales a lo largo del periodo de elución del análisis hplc para los compuestos fenólicos individuales, y omitiendo así el innecesario proceso espectrofotométrico. para la validación del método propuesto, se estableció un sistema modelo de solución de vino, compuesto de cianidin- 3-o-glucósido, epicatequina y acetaldehído con una proporción de 1: 6: 10 (polifenoles totales 4,366 mg/ml), etanol al 12%, 5g/l de ácido tartárico y a ph 3,2.durante la reacción cinética en la solución de vino modelo, que simula el proceso real de almacenamiento y envejecimiento del vino, se controlaron periódicamente los polifenoles totales mediante el método del índice de riberaux-gayon (absorbancia a 280 nm) y el método del índice de folin-ciocalteu, así como el método propuesto de integración de picos. los resultados han mostrado que no había diferencias significativas en el contenido total de polifenoles entre los tres métodos (p>0,05), presentando todos ellos una buena precisión en comparación con el contenido conocido de polifenoles en la solución de vino modelo. además, la viabilidad del método de integración de picos propuesto también se verificó mediante su aplicación a la cuantificación de polifenoles totales en varios vinos tintos diferentes. los resultados obtenidos por el método de integración de picos propuesto fueron bien consistentes con los obtenidos por el índice de riberaux-gayon y el índice de folin- ciocalteu (p>0,05), indicando que el método propuesto es válido para la determinación de polifenoles totales en vino tinto.

Validation of a method for the determination of volatile compounds in in spirituous beverages using contained ethanol as a reference substance
